Electronic Pipettes

An electronic pipette uses a motor instead of your thumb to move the piston. Liquid handling, explained without the jargon

Written for whoever is actually raising the purchase request — a new analyst, a lab manager, a procurement officer. No prior knowledge assumed, no marketing language.

What is a pipette?

A pipette is a calibrated instrument that measures and transfers a precise volume of liquid, usually between 0.1 microlitres and 10 millilitres. Think of it as a syringe with a certified accuracy specification. The volume it delivers is traceable, verifiable and central to whether your result is defensible.

How a pipette works

Most laboratory pipettes are air-displacement. A piston moves up inside the shaft, creating a small vacuum that pulls liquid into the disposable tip. An air cushion sits between the piston and the liquid — the liquid never touches the pipette itself. Press to the first stop to set the volume, release to aspirate, then press through to the second stop to expel everything including the residual film.

Mechanical vs electronic pipettes

Mechanical pipettes depend on your thumb: how fast you press, how long you pause, how hard you push. Electronic pipettes replace the thumb with a motor, so speed and stroke are identical every cycle. Mechanical wins on cost, simplicity and no charging. Electronic wins on repeatability, ergonomics and programmable modes. A practical rule: below two hours of pipetting a day, mechanical is fine; above that, the injury risk and the operator variance both argue for electronic.

Single channel vs multichannel

A single channel handles one tube at a time. A multichannel carries 8, 12 or 16 tips at the standard microplate spacing so you fill a whole row or column at once. If you work with plates you need multichannel — a 96-well plate takes 96 actions with a single channel and 8 with a 12-channel.

How to choose the right volume range

Accuracy is worst at the bottom of a pipette's range. A 100–1000 µL pipette set to 100 µL is far less accurate than a 20–200 µL pipette set to the same 100 µL. Choose the pipette whose nominal (maximum) volume is closest to and above your typical working volume, and try to operate in the upper 35–100% of the range. Never set a pipette outside its stated range — it damages the mechanism and voids the calibration.

How pipette tips work

The tip is a disposable moulded plastic cone that seals against the pipette shaft. That seal is everything: a poor fit lets air leak past, and the volume you deliver is smaller than the volume you set — silently, with no error message. Tip fit, plastic quality and moulding consistency directly determine whether your calibrated pipette actually delivers its specification.

Standard tips

Plain tips with no filter and no sterility claim. Correct for the majority of routine work: adding buffers, transferring reagents, making dilutions where nothing precious or hazardous is involved. The cheapest option and, for most benches, the right default for the bulk of pipetting.

Filter tips

A porous plug in the top of the tip stops aerosols and liquid from reaching the pipette shaft. Two things get protected: your instrument from contamination, and your next sample from anything left behind by the previous one. Mandatory for PCR, patient samples and anything infectious, radioactive or corrosive.

Sterile tips

Sterile tips are gamma-irradiated and sealed so nothing viable is present when you open the rack. Required whenever contamination would ruin the work — cell culture, sterility testing, microbiology. Note that sterile and nuclease-free are different claims: a sterile tip is not automatically DNase or RNase free.

Biopur and certified purity tips

The highest certification grade. Each lot is tested and documented free of DNA, DNase, RNase, PCR inhibitors, ATP and endotoxins. You pay more per tip and in return you eliminate the consumable as a suspect when a sensitive assay behaves unexpectedly. Worth it for qPCR, NGS, forensic DNA and any assay where a repeat run costs more than a case of tips.

ClipTip technology

Conventional tips are held on by friction, which means pressing hard and hoping every tip on a multichannel head seated equally. ClipTip tips mechanically clip into place instead. The practical benefit is uniform seating across all channels — lower channel-to-channel variation on plate work — and much less force on your hand and wrist. The trade-off is a closed system: ClipTip tips work on ClipTip pipettes, not universal ones.

Bottle-top dispensers

A dispenser that mounts on the reagent bottle itself and delivers a set volume with one stroke. It exists mainly for safety: you stop pouring concentrated acids and solvents into open glassware. A recirculation valve returns the priming volume to the bottle instead of wasting it. Choose wetted materials — borosilicate, PTFE, ETFE — to match your reagent chemistry.

Repeat dispensers

A positive-displacement device that fills one syringe tip and dispenses it in up to a hundred equal steps. Two reasons to own one: speed, when you are filling plates or aliquoting reagents, and accuracy on liquids that defeat air-displacement pipettes — glycerol, serum, oils, volatile solvents and anything that foams.

Serological pipettes

Long graduated pipettes for millilitre volumes, 1 mL to 100 mL, used with a controller. Plastic disposables come sterile and individually wrapped for cell culture; glass versions are autoclavable and preferred for solvents and long-term economy. Colour-coded volume bands let you grab the right one without reading the barrel.

Pipette controllers

The motorised handle that drives a serological pipette. Two-speed control matters more than it sounds: fast for media changes, slow and gentle for resuspending cells you do not want to shear. The hydrophobic membrane filter is a consumable — replace it when aspiration slows or if liquid ever reaches it.

Calibration

Calibration is gravimetric: dispense water onto an analytical balance, convert mass to volume using the density of water at the measured temperature, and compare against the manufacturer's accuracy and precision specification. Standard practice is external certified calibration every 6 to 12 months, with internal quick checks quarterly or after any drop, service or change in the type of liquid handled. For accredited labs, ISO 8655 is the reference method and the certificates form part of your audit trail.

Maintenance

Daily: wipe the exterior and store the pipette vertically in a stand — never lying on the bench where the tip cone gets knocked. Weekly: check the tip cone for scratches and residue. Quarterly: inspect and grease the piston, replace O-rings and seals. A leaking seal is the most common cause of a pipette that suddenly under-delivers, and a maintenance kit fixes it on the bench in minutes. Always store a pipette at its maximum volume setting to relieve spring tension.

Which pipette is best for HPLC sample preparation?

For HPLC vial filling, a manual single-channel variable pipette in the 100–1000 µL range covers most sample and diluent transfers, with a 10–100 µL unit for standards and spiking. Use extended-length tips so the pipette barrel stays clear of the vial neck. If you prepare large batches of mobile phase or diluent, add a bottle-top dispenser on the solvent bottle — it is faster and considerably safer than pouring.

Which pipette is best for PCR?

Use a dedicated low-volume set — typically 0.1–2.5 µL, 0.5–10 µL, 2–20 µL and 20–200 µL — reserved exclusively for PCR work and never taken outside the pre-PCR zone. Filter or barrier tips are non-negotiable. For plate-based setup, an 8 or 12 channel electronic pipette cuts both time and pipetting variance across the plate.

Mechanical or electronic — how do I decide?

Decide on pipetting hours and repeatability requirements, not budget alone. Under two hours a day with varied protocols: mechanical. Over two hours a day, or repetitive plate work, or a validated method where operator variance must be controlled: electronic. Most labs run a mixed fleet — mechanical as the bench default, electronic where throughput and ergonomics justify it.

What volume range should I choose?

Match the pipette's nominal volume to just above your usual working volume and operate in the upper part of the range. Accuracy degrades sharply at the bottom of any range. Dispensing 50 µL is more accurate on a 10–100 µL pipette than on a 100–1000 µL pipette, even though both can technically reach it.

How many pipettes does a new laboratory actually need?

A workable starting set per bench position is four manual single channels — 0.5–10 µL, 10–100 µL, 20–200 µL and 100–1000 µL — which covers roughly 90% of routine work. Add a 12-channel if you run plates, a bottle-top dispenser if you handle bulk reagents, and a repeat dispenser if you aliquot frequently. Should I standardise on one brand across the lab?

Standardising simplifies tip purchasing, calibration contracts, spares and training — real operational savings. The exception is a closed tip system like ClipTip, which locks you to a matching tip supply. Many labs standardise on one universal-fit brand for general benches and run a closed system only where its specific benefit applies.

Fixed volume or variable volume?

Variable volume covers a range and suits changing protocols — the right default for most labs. Fixed volume repeats one value only, which removes the risk of a mis-set dial and speeds up highly repetitive routine tests. Consider fixed volume for a single high-frequency validated step, variable for everything else.

Do I need a positive displacement system?

Yes, if you handle liquids where the air cushion in a normal pipette introduces error: viscous liquids like glycerol and serum, volatile solvents, dense liquids, and foaming solutions. A repeat dispenser or positive-displacement pipette puts the piston in direct contact with the liquid and removes that error source.

Which pipette tip is compatible with my pipette?

Compatibility depends on the tip cone geometry of your specific model. Universal-fit tips cover most major brands, but closed systems like ClipTip only accept their own tips. What is a filter tip?

A filter tip has a porous plug near the top that blocks aerosols and liquid from entering the pipette shaft. It protects the instrument from contamination and prevents carry-over into the next sample. Standard practice for PCR, clinical samples and any hazardous liquid.

What is an ART barrier tip?

ART barrier tips add a self-sealing barrier that physically seals on contact with liquid, rather than only filtering aerosol. If a user over-aspirates, the barrier closes and nothing reaches the pipette. Widely used in nucleic acid amplification and clinical laboratories where cross-contamination would invalidate results.

What is a Biopur tip?

Biopur is Eppendorf's highest purity certification. Each lot is tested and documented free of DNA, DNase, RNase, PCR inhibitors, ATP and endotoxins. Used where consumable purity must be excluded as a variable — qPCR, NGS, forensics and sensitive cell-based assays.

What is ClipTip?

ClipTip is Thermo Scientific's mechanical tip attachment system. Tips clip onto the pipette instead of being held by friction, so every channel on a multichannel seats identically and attachment force is much lower. The benefit is lower channel-to-channel variation and less strain on the hand; the constraint is that ClipTip tips fit only ClipTip pipettes.

When should I use low-retention tips?

Whenever liquid clinging to the tip wall costs you money or accuracy — detergent-containing buffers, protein and enzyme solutions, master mixes, and any sample you cannot afford to lose. The hydrophobic surface releases more of the liquid, improving recovery and reproducibility.

What are wide bore tips used for?

Wide bore tips have a larger opening so shear-sensitive material passes through undamaged. Use them for cell suspensions, genomic DNA, viscous samples and anything containing particles that would be sheared or blocked by a standard tip orifice.

Are sterile tips the same as nuclease-free tips?

No, and this catches people out. Sterile means no viable microorganisms, achieved by gamma irradiation. Nuclease-free means no DNase or RNase activity, which is a separate test and certification. Molecular biology work usually needs both — check that the certificate states each claim explicitly.

Can I autoclave pipette tips?

Many tips can be autoclaved at 121°C for 20 minutes, but check the manufacturer's statement first. Autoclaving destroys the filter in most filter tips and does not make a tip nuclease-free — heat does not degrade DNA contamination. For nuclease-sensitive work, buy certified tips rather than autoclaving standard ones.

What is the difference between racked, reload and bulk tips?

Racked tips come upright in a rigid rack — fastest and most hygienic, and the standard for sterile work. Reload tips are stacked layers that drop into a rack you already own, cutting plastic and cost. Bulk tips come loose in a bag at the lowest cost per tip, suitable for non-sterile routine work. Many labs use racked for sensitive work and bulk for general pipetting.

Do cheap unbranded tips actually affect my results?

Yes, more than most buyers expect. Inconsistent moulding causes variable sealing on the tip cone, which means variable volume delivery from an otherwise calibrated pipette. Plastic additives can leach and interfere with assays. If you are spending on a certified pipette and then fitting an uncontrolled tip, you have given away the specification you paid for.

What are gel loading tips?

Very fine, flat and flexible tips that fit into electrophoresis gel wells. They let you deliver a sample into the well without piercing the gel matrix or contaminating adjacent lanes. Available in round and flat variants for different well geometries.

How often should pipettes be calibrated?

External certified calibration every 6 to 12 months is standard, with the interval depending on usage intensity and your quality system. Regulated laboratories — pharmaceutical QC, NABL-accredited diagnostics — usually run 6-month cycles. Add an internal gravimetric check quarterly and always after a drop, a repair, or a change to a significantly different liquid type.

How is a pipette calibrated?

Gravimetrically, per ISO 8655. You dispense distilled water onto a calibrated analytical balance, record the mass, and convert to volume using the density of water at the measured temperature and pressure. Ten replicates at minimum, mid and nominal volume produce the accuracy and precision figures that are compared against specification.

Can I calibrate pipettes in-house?

You can perform verification checks in-house with a suitable balance and controlled conditions, and many labs do this quarterly. Certified calibration for audit purposes should come from an accredited laboratory that issues a traceable certificate. What does ISO 8655 cover?

ISO 8655 is the international standard for piston-operated volumetric apparatus. It defines maximum permissible errors for accuracy and precision by volume, and specifies the gravimetric test method, environmental conditions and number of replicates. It is the reference document that pipette specifications and calibration certificates are written against.

Why is my pipette suddenly under-delivering?

The most common cause is a worn or damaged seal or O-ring letting air past the piston. Next most common is a scratched or contaminated tip cone giving a poor tip seal, followed by incorrect technique — inconsistent immersion depth or aspiration speed. A maintenance kit resolves the first two on the bench in minutes.

How should pipettes be stored?

Vertically, in a stand or carousel, at the maximum volume setting. Vertical storage protects the tip cone from bench damage and stops residual liquid running back into the shaft. Setting maximum volume relieves spring tension and extends the working life of the mechanism.

Can pipettes be autoclaved?

Many manual pipettes have a fully autoclavable lower assembly — check your model's manual, because the specification varies. Electronic pipettes have autoclavable lower parts but the electronics never go into an autoclave. Allow the assembly to dry and cool fully before reassembly, and recalibrate after a series of autoclave cycles.

What is in a pipette maintenance kit?

Typically O-rings and seals for the relevant volume range, piston grease, a service tool, and sometimes a replacement piston or shaft. It lets your team fix the most common failures without sending units away, which matters when a pipette is on a validated method and downtime stops testing.

How long does a laboratory pipette last?

A well-maintained quality pipette commonly stays in service 8 to 10 years or more, with periodic seal replacement and calibration. Failure is usually a serviceable part rather than the instrument. Electronic units are additionally limited by battery life, which is normally replaceable.
